Solution

The pLysS plasmid encodes for T7 lysozyme. This enzyme is produced to keep the T7 RNA polymerase (which is encoded by the DE3 region of the host chromosome) in check. The T7 lysozyme binds to T7 RNA polymerase and inhibits it, reducing the basal expression level of the T7 RNA polymerase prior to induction. This is particularly useful when expressing toxic proteins.
